NAD+ (nicotinamide adenine dinucleotide) is the central redox coenzyme of cellular energy metabolism and a substrate for the sirtuin and PARP enzyme families; tissue levels decline with age. 5-Amino-1MQ is a selective inhibitor of nicotinamide N-methyltransferase (NNMT); blocking NNMT spares nicotinamide for NAD+ synthesis via the salvage pathway and has been associated with reduced adipose mass in animal models. MOTS-c is a mitochondrial-derived peptide encoded in the 12S rRNA region of mitochondrial DNA, reported to activate AMPK and improve metabolic flexibility. None of the three acts on the growth-hormone / IGF-1 axis. Human trial data remain limited for 5-Amino-1MQ and MOTS-c.
